It is a molecule naturally produced by the body that occurs in all living cells. The plus in the name refers to the oxidized form. When NAD+ accepts electrons, NADH is formed. By constantly switching between NAD+ and NADH, this redox pair helps make energy from carbohydrates, fats, and proteins available.<\/p>\n<p>NAD+ has two main functions:<\/p>\n<ol>\n<li><strong>Coenzyme in energy metabolism.<\/strong> NAD+ is involved in, among other things, glycolysis, the citric acid cycle, and energy production in mitochondria. It is not the energy source itself, but helps reactions proceed that ultimately contribute to the formation of ATP.<\/li>\n<li><strong>Raw material for NAD+-consuming enzymes.<\/strong> Among others, sirtuins, PARP enzymes, and CD38 use NAD+ in processes surrounding cell signaling, stress response, and DNA repair. In the process, NAD+ is consumed and breakdown products are created that the body can partially recycle.<\/li>\n<\/ol>\n<p>Therefore, it is not just about &#8220;how much NAD+&#8221; is present. The NAD+\/NADH ratio, the location in the cell, and the speed at which NAD+ is produced and consumed are also important.<\/p>\n<h2 id=\"waarom-daalt-nad\">Why can NAD+ decline as you get older?<\/h2>\n<p><strong>The most accurate answer is that the NAD+ balance can shift with age.<\/strong> Research describes a decline in various animal models and in certain human tissues. At the same time, evidence in humans is still limited and tissue-specific. A large study from 2026, for example, found no age-related decline of NAD+ in whole blood. In preclinical research, these signals stimulated CD38-positive immune cells, resulting in lower NAD+ availability.<\/li>\n<\/ul>\n<p>There is therefore no scientifically substantiated universal rule that everyone at age 40 only has 25% of their original NAD+ left. It is an intermediate product in the production of NAD+. In cells, NMNAT enzymes can convert NMN into NAD+.<\/p>\n<p>The simplified pathway is:<\/p>\n<p><strong>nicotinamide \u2192 NMN \u2192 NAD+<\/strong><\/p>\n<p>After oral intake, NMN is absorbed and metabolized. The exact absorption likely differs per intestinal segment and tissue. A portion may first be converted outside the cell into nicotinamide riboside (NR), enter the cell, and form NMN there again. Direct uptake of intact NMN has been proposed in some models, but the mechanism and its importance in humans are still subjects of research.<\/p>\n<p>Therefore, &#8220;NMN goes directly into every cell&#8221; is too definitive.
