Comparing NAD+ supplements: NMN, NR, niacin, NADH, and NAD+
Short answer: NMN and NR (nicotinamide riboside) are the best-researched NAD+ boosters: in human studies, 1 gram per day doubles the NAD+ levels in the blood within about two weeks. Niacin (nicotinic acid) also works strongly but often causes a flush at higher doses. Nicotinamide increases NAD+ only briefly. For NADH and oral NAD+, there is little evidence that they are absorbed intact. NMN vs NR: what do direct comparisons say?
In 2026, two studies were published that directly compared NMN and NR:
- Christen et al., Nature Metabolism 2026: 65 healthy adults received 1 g NR, 1 g NMN, 0.5 g nicotinamide, or placebo for 14 days. NR and NMN approximately doubled NAD+ in the blood; the difference between the two was not significant. Nicotinamide only gave a short peak. Notably: NR and NMN were mainly converted by gut bacteria into nicotinic acid, which the body then uses to make NAD+.
- Berven et al., iScience 2026: in a small study (1,200 mg/day), NR resulted in approximately 2.3 times more increase in blood NAD+ than NMN. After four weeks, NAD+ also increased in the brain, by about 35% in healthy participants.

Why NADH and oral NAD+ are less logical
NAD+ and NADH are relatively large molecules. After ingestion, they are largely broken down in the gut into smaller building blocks such as nicotinamide, NMN, and NR. There are few human studies showing that oral NAD+ or NADH increases NAD+ in cells better than the precursors themselves. A 2023 systematic review (American Journal of Physiology โ Endocrinology and Metabolism) concluded that the evidence for NAD+ administration in various conditions is limited and of varying quality. NAD+ infusions do increase levels directly, but they are expensive, take hours, and have hardly been studied for long-term effects.
Which NAD+ supplements are independently tested and safe?
