Looking for the best supplements for energy and focus? Those who are structurally tired or have difficulty concentrating often look to supplements. However, not every “energy supplement” targets the same cause. Vitamin B12, B6, and folic acid have an EFSA-recognized, scientifically evidence-based function in normal energy-yielding metabolism and the reduction of tiredness and fatigue in cases of deficiency. Magnesium supports muscle and nerve function and, according to recent research, may play a role in cognitive performance. NMN, GlyNAC, and TMG target deeper cellular processes such as NAD+ production, mitochondrial function, and methylation — with varying degrees of evidence. Why do you feel tired and unfocused?

Fatigue and concentration problems rarely have a single cause. Sleep deprivation, chronic stress, a deficiency in iron, vitamin B12, or vitamin D, insufficient calorie or protein intake, and underlying medical conditions (such as thyroid issues) are all common causes that are separate from “metabolic balance” in the sense that supplement brands use the term. Before adding a supplement, it is sensible to first rule out these root causes — possibly with a blood test via your GP.

Only once nutrition, sleep, and exercise are in order does it make sense to look at supplements that support energy metabolism at the cellular level.

What do we mean by “metabolic balance”?

Metabolic balance refers to how efficiently your body converts nutrients into usable energy (ATP) via the mitochondria, and how stable processes such as blood sugar regulation, homocysteine metabolism, and oxidative stress balance function. Supplements that address this do so via different routes: some fill a direct deficiency (vitamins, minerals), while others influence underlying cellular processes (NAD+, glutathione, methylation).

Supplements for energy and focus with the most evidence-based effect

For the following nutrients, energy-related claims have been assessed and permitted by the EFSA under Regulation (EC) 1924/2006 — provided there is actually an insufficient intake:

  • Vitamin B12: contributes to normal energy-yielding metabolism and the reduction of tiredness and fatigue; relevant in case of a deficiency, which occurs more often with plant-based diets or in the elderly.
  • Vitamin B6: same recognized claims, with an additional role in homocysteine metabolism.
  • Niacin (B3): contributes to normal energy-yielding metabolism, normal psychological functions, and the reduction of tiredness and fatigue.
  • Folic acid: supports homocysteine metabolism along with B12 and B6, which is indirectly related to cellular energy.

The most important caveat: these effects have been demonstrated when addressing a demonstrable deficiency, not as a general “boost” with an already adequate intake.

Magnesium: potentially relevant for focus, not just for energy

Magnesium is involved in more than 300 enzymatic processes, including energy production. A systematic review from 2025 confirms the role of magnesium in muscle, nerve, and energy metabolism, and indicates that a significant portion of the population does not meet the recommended intake. More interesting for “focus” specifically is a randomized, double-blind, placebo-controlled trial from 2025 on magnesium L-threonate (Magtein®), which showed improvements in cognitive performance and sleep quality in adults. This is one study with a specific form of magnesium — not a confirmation that all magnesium supplements have the same effect, but a concrete indication that magnesium plays a role beyond just “no cramps.”

NMN, GlyNAC, and TMG: where do they fit into this picture?

These three target the cellular level more deeply than filling a vitamin or mineral deficiency, and the level of evidence clearly differs per substance:

GlyNAC (glycine + N-acetylcysteine) has the strongest evidence-based effect of the three on functional outcomes. A randomized clinical trial in older adults (Kumar et al., Houston Methodist/Baylor College of Medicine) showed improvements in glutathione levels, mitochondrial function, muscle strength, and cognition after 24 weeks of supplementation. Note: this has been demonstrated in older adults with a demonstrable glutathione deficiency, and is not necessarily generalizable to healthy younger adults.

TMG (trimethylglycine/betaine) has a well-documented, defined function in homocysteine metabolism via the betaine-homocysteine methyltransferase pathway, which is relevant for methylation — but this is not the same as a direct effect on energy or focus.   1. First, rule out non-supplement-related causes (sleep, iron, thyroid, stress) — in case of persistent fatigue, consult a physician.
  2. Start with a demonstrable deficiency: a blood test shows whether B12, vitamin D, iron, or magnesium levels are actually low.
  3. Distinguish “biomarker improves” from “I feel noticeably better” — for NMN, this distinction is crucial, for example.
  4. Do not combine multiple supplements without consultation, especially not when using medication, during pregnancy, or with underlying conditions.
  5. Give a supplement a realistic trial period (usually 4–12 weeks) before assessing the effect.
